The Emperor was at some point capable of creating babies, but apparently they were either wiped out or driven into exile during the Age of Apostasy, so they're not around anymore. Also, the Primarchs are essentially lesser clones of his, so I don't think they have a whole lot of urges.

The real question should be: Can the Emperor pass on all of his power genetically? Or does he only possess all of his power because of the sacrifices made in the past as well as the current sacrifices made in his name?

Could someone recreate the Emperor in everything but memories/personal experience by using a sample of his DNA to create a clone of him via a surrogate mother/test tube incubator?

The emperor had kids. He can do what he wants. Primarchs and space marines can't have kids. They are sterile because the emperor did not intend them to replace regular baseline humanity. He wanted baseline humanity protected and to evolve into mini-emperors themselves. That's why he clings to life. If he can survive long enough for that to happen, humanity wins.

The primarchs and space marines were designed to guide and protect humanity, not replace it. And the ability to breed would be too much of a risk to that goal, so that's why their only (current) way to reproduce is recruitment from people. Corax and the Emperor (and not Roboute with the primaris marines) was able to just vat grow them and make them mature quickly. While for the last 10,000 years everyone was stuck with the jerry rigged and inefficient backup way which was mainly to serve as a way for them to recruit on the go during the great crusade rather than wait for recruits to be made before being able to replenish their ranks.
