I'd honestly love it if there were an super difficulty level where going into Zot locks you in forever, and you have to complete a Ziggurat (which would contain the Orb) to get out.

Flavor-wise, the Ziggurat could be a tower going up. So, since the Dungeon+Depths+Zot is 25 floors and Zigs are 27, at the end of a Zig, you'd take the Orb to Heaven.

No.

The most dangerous rune for MuMo is probably Gloorx because of speed 20 DU spam.

As far as hell lords go, Asmodeus requires decent itemgen to have enough rF if you want to manfight, but otherwise shouldn't be a problem assuming that the Geh:7 floor generator doesn't spawn uncrossable terrain. Ereshkigal is more or less trivial as long as you fight her solo. Antaeus hits like a truck and would be dangerous, but doable, again though the problem is the floor layout because boots of flying are for elves. Dispater might be problematic if your gear drops have been subpar, but is more or less on par with Antaeus. Mind, you can ninja all of these if you've saved enough scrolls, and doing so is definitely the smart way to go about it.

Coc is near guaranteed to be not-doable without flight. Geh is roughly the same. Dis and Tar are passable.

Lightning spire. Animate undead. Deflect missiles. Haste, if its still in the game. Regeneration. If you want to get cheeky, OTR, Pcloud, Parrow and Ignite poison for maximum fun. Pcloud woulds extremely well with animate undead tho, so its worth a pickup anyway.

the AC bonus is priceless, no-breathing is high utility.
Grey draconian with dragonform is wild because it counteracts the one thing dragonform sucks with- defense.
