I assume you're not talking about adding an obvious parody character.
For a transsexual character, create a character that is mostly feminine but has a few 'off' masculine features (height, slightly broader shoulders, narrow hips etc.) but nothing that is extremely obvious. The character should have the near universal transsexual traits like shyness, low-self esteem, overly feminine dress, makeup, hairstyle (to compensate for male features) and aversion to personal questions and relationships. You reveal that the character is trans in a situation like, someone is interested in the character romantically and they're forced to reveal their status to them, someone walks in on them changing, a third party character who knew them as a male before transition sees them and blurts out the information, things like that. The key to realism is that it shouldn't be obvious that they're trans, but once you actually discover they're trans, it should feel like it "should have been" obvious. The signs should be hidden but still there.
Futanari, you'll need to go the Ninier route. A character that was cloned or created and 'mistakenly' given male genitalia, a character that was accidentally changed by a misfired potion or spell or machine, a character that was 'cursed' with male genitals by an adversary, things like that.
Actual real life intersex people with genitals like the opposite sex are never attractive. In reality the genitals appear deformed or 'ambiguous'. It's hard to sell a character that is a girl that 'just so happened to be born with a penis' because that's not how it works in reality.

Depending on what you're hoping to get out of adding a trans character. If you want fanart bait, it may be wiser to add all of the characteristics mentioned, but leave it ambiguous. Of course this works better in games that are more mechanically based and not as story-focused, you can get away with telling less.

rougebasin articles. Start with the basic room, corridor, approach or perlin noise caves, then expand from there. Rooms aren't rectangles/squares but one of a list of shapes, so on.
Mostly, just play rougelikes, every dev has a devblog. The community is pretty open, etc.

You're still learning so, while 8 hours is objectively terrible for that result, it's fine.
But you may find it helpful to set a time limit for practice. 1 hour to sketch the design, 1 hour to polish, once you're done you're done. Make sure after you're done you stop and look at what you did wrong or right. You could have made 4 sprites in that same time!

But what will probably happen is as you get better, you won't keep making animations like that, the quality will improve while the time you take per sprite remains roughly constant (unless you're aiming to get shit done fast)

Introduce movement. Maybe a jetpack that launches you a set distance up and forward (so you're not always going backwards) or dark soul rolls with I frames. You could also try adding a "base", that way you're not always going backwards but instead around?
I accidentally closed the itch tab which contained your written feedback, but the gist was that the plugin system is a good idea for the type of game you're going for, since it felt that one could run out of stuff to do rather fast. And I always ask whether you want to add a plugin creator for users. I liked that game, although I may be a bit biased because the zerg rush stuff reminds me of Mirklings.
