I'm assuming this excludes Gil, as there's no fight otherwise. It really comes down to Cu and Artoria.
Artoria's biggest flaw is how she's ALWAYS underestimating her opponent and overestimating herself. If she can leave that behind and start fighting seriously from the beginning, she stands a much better chance. She'd have to stop her "must hide identity" bullshit. That always hurts her more than anything. She's always been able to fight better after revealing her identity, while hiding it always puts her at a disadvantage. Fix all of these, make her seriously focused only one this fight and nothing else, and you have a strong Servant. Even give her Avalon.
Cu, on the other hand, has consistently showed great results. He's pretty eager in using his NP, and that NP is pretty great. It allows him to deal an unavoidable blow at close range, and he can even use it at long range. He can fight at close range amazingly well without using his NP, and has a skill that lets him block long ranged attacks. Battle Continuation allows him to just not die. By all accounts, he'd win.
Avalon helps Artoria a bit. As a defense, Avalon is pretty useless. In battle against Gil, she had to survive being hit by Ea multiple times just to get the timing right to use Avalon. Gil only let her live because, well, he didn't want her dead. Cu doesn't care. He'll go for the kill, and Avalon isn't much use defensively in that case. Its healing powers WILL let her survive one Gay Bulge easily, though, so there's that.
The BEST chance Artoria has is to Excaliblast him before he can put up a fight. Even then, since the beam is kind of a projectile, and since Cu is really fucking fast, he might be able to avoid it. Even DUDE and Gil could easily avoid it, Cu has a better chance than those two. And he has Battle Continuation on top of that. He's going to get in a few hits, there's not much Artoria can do to avoid that. It comes down to a battle of attrition, and Cu doesn't die.
