Well, autism issue aside, it's all about being able to get into a character, or at the very least, to know them well. A lot of it is the language they use, too.

Just to go with a simple example...
If someone asks the main girls (misha included, rin excluded) a simple question as 'how are you doing?' and (removing [sign language], s-stuttering and stuff like that) you get these 5 answers :
>Great, great!
>I'm good!
>I am well, how are you?
>I'm fine!
>I'm ok.
Would you be able to figure out who said what? If you aren't, is it because you don't know the characters well enough, or because you think some of these lines would fit 2+ characters equally well? Emphasis here is on "equally" because of course they could all say these 5 lines, but (imho) they are NOT as likely to say them, and even if some are very close, the nuance between the characters makes the difference.

and for the record, it's not necessarily about getting the 5 right (there might not be a right/wrong anyway) it's about being able to figure out your interpretation and justify it with ingame/character thing, because that's what shows you get the character right.
What I mean by that is that, while I had 5 specific names in mind when I wrote the 5 replies... You could have say Emi go with "I'm ok" because she doesn't want to talk about something, or with "I'm good!" to hide better the fact that she's in fact not good, or even with the more lilly-like "I am well, how are you?" because you think Emi would change the subject to avoid you pressing further on whatever answer she gave you;
It's mostly about putting yourself in the character's head, getting it right, his/her motivations/habits and all that.

That 1 line of dialogue is obviously a bit vague, but to put it this way : If someone was to read a 20'ish lines dialogue between 2 characters with blanked names and he can't figure out who they are, they're probably not written in character.

The language (level of language, specific words/expressions they use, etc..) should be a big clue, and also the dialogue should reflect the attitude of the character.

Say, just something as simple as

This doesn't sound like Hanako, or Lilly (she's probably come up with something better than just 'great'), it's too enthusiastic for Shizune or Rin, so it's a bit more like Emi, and A LOT more like Misha (barring me changing the Hisao not being "hicchan", for obvious reason).
While Emi is also very enthusiastic like Misha, this line doesn't sound as much like her to me as it does Misha.
I would see Emi more like "It's great!" or just "Great!", and not necessarily saying "Hisao" at all.

Not sure if any of this made sense to you, but tht's how I see it.

Na, I mostly lurk. First survey I've done; I was just curious if there was a correlation between favorite katawa and first route or first impression. Currently seems like first route is stronger, but not particularly influential.

as for me ( ) I knew after meeting the 5 girls that Emi would be my best, so I kept her for last. I had a pretty good idea where they all would stand too, before the path-locking decisions. I wanted to go blind the first time, but after that I knew who I'd like/wouldn't like, and was spot on on everyone but Rin. I mean I knew I'd like her, but I was wrong on how I pictured her I guess. But everyone else was kinda easy to figure out imho.

yeah, I personally like Rin better in Emi's path than in her own path. If Rin had always been like act 1 Rin (with some other drama, or maybe just a lesser version of that one she got) she might have made a push for best girl(to me). But she's a bit too much in her path.
