The Island. If possible build a large bridge between it and the mainland. It's easily defensible and can act as a port. Besides that it's nice and flat.

At Quest Point

Build between the northernmost riverfork as it reaches the sea. We'll have a port, excellent farmland, and nearby mountains for mines and as a natural defensive wall.

Ah, well, unfortunately as far as we can tell, the island is entirely barren. There's no water anywhere to be found, and the soil is full of salt. About a day's journey south from the Northernmost point there's a large hole, leading into an underground cavern, but we were unable to explore it to any appreciable extent for fear of running low on supplies.

If his majesty wishes, we might be able to build some sort of structure on the island once the region proper is more fully developed?

Get some sort of defensive wall set up. Then a port.

Why not the southwest corner then? Too small?

Assuming the capital or a current city is the north, right between the mountain and the river on the upper right. Good defense, the river can be fished and used to float product. Begin constructing a tunnel or path through the mountains so the new found colony has an easy path to the kingdom lest they go rouge in such a bounty.

Enslave goblins and then release them on the colony's doorstep in the middle of the night

Send some of our main colony down there to destroy the goblins and take over the southern settlement. Still make the northern settlement that the other guy suggested.
And, if possible, send some colonists to the area in the left-middle, between the river and the mountains, next to the Great Lake.

Excellent analysis. Most of the initial lumber was used in construction of the buildings, but ship construction will begin in short order.

Establishing colonies up river will be dangerous, and the journey treacherous, as it would have to be made on foot. Perhaps sending scouts would be of use?

A path has been cleared to the mountains, and a quarry has been constructed. Once sufficient stone has been extracted, we'll begin construction of more defensible structures.

Now that the initial settlement has been established, we should look to gaining a foothold in more regions of the area. Should we send colonists South, down the Skargrave river to establish colonies within the heart of the region, or farther along the coastline?

Both scouting parties have returned now majesty.

The Eastern scouting party went as far as the Eastern tip of Great Lake. The water there is clear and fresh, with clay filled banks. The clay seems to possess some strange properties, namely that it is always cool to the touch, even after being pulled from a hot flame.

The lake is home to a dazzling variety of fish, many of which taste delicious. It is also home to a terrifying race of sapphire serpents, gigantic beasts capable of swallowing a deer whole. They attacked the party only once, contenting themselves to hunt the fish in the lake for the most part.

The Southern scouting party stopped at each of the deltas. They found plentiful game, including a strange species of scaled, bipedal lizard which ate leaves. It moved by hopping, and came up about waist high. Their meat is quite tender, and they are plentiful in that area.

As they traveled along the Southern area, they found many tribes of ogres. This most horrible race of men had green, dappled skin all across their bodies, and wore almost no clothing save a loincloth. For weapons, they wielded entire trees, for they have no fire and eat their meat raw. The men kept a large distance, for fear of being consumed, for anytime one of the brutes caught sight of a man they roared and gave chase. Given their incredible bulk, they were quite easy to outrun, but if the area is to be settled they must be dealt with eventually.

Set up that fort in the northwest, and get an army and colonists ready to really take the area. Strengthen our First Colony, and build homes to be promised cheaply to colonists.
